DETECTION AND QUANTIFICATION OF Pseudomonas syringae PV. lachrymans IN CUCUMBER SEEDS LOTS

Detection of Pseudomonas syringae pv. lachrymans was made by direct planting. After germination, typical leaf spots were submitted to pathogenicity tests. Percentage of transmission varied from 1,0 to 2,5%. A selective medium for Pseudomonas syringae pv. lachrymans was thought out by adding to the medium 523 of Kado & Heskett 200g/ml of bacitracin, captan, lincomicin and bengal rose. Seeds were deposited into the surface of the selective media and colonies showed up were counted and tested for pathogenicity. This method was able to detect bacteria in 1 seed lot out of five. For quantifleation 500 seeds were submitted to extraction in PBS containing antimicrobial compounds cited. Seed extracts were dilueted for plate counting. Typical, single colonies were tested for pathogenicity. This method allowed detection of P. syringae pv. lachryrnans in all seed lots and UFC values per gram of seed varied from 1.5 x 107 to 2.5 x 105.
