DIFFERENTIATION OF Colletotrichum gossypii AND C. gossypii var. cephalosporioides BY ISOENZYMES IN COTTON SEEDS

The differentiation of Colletotrichum gossypii e C. gossypii var. cephalosporioides by
electrophoretic analysis using isoenzymes was investigated in pure culture. The isoenzyme analysis was
performed in electrophoresis 1D-PAGE at 7,5% (separating gel) and at 3,5% (concentrating gel), using
Tris-Glycine pH 8,9 as standard buffer in the gel-electrode system. Out of the five isoenzymes systems
tested, the profiles of esterase, phosphatase acid and peroxidase were able to distinguish the isolates of both
Colletotrichum in the present work. The results of the pathogenicity and electrophoresis assays were correlated
which makes this technique as a promissing tool to be used to differentiate both pathogens
