GENETIC SIMILARITY AMONG CARIOCA BEAN CULTIVARS BY MOLECULAR MARKERS OF PROTEIN AND ENZYME FOR GENETIC PURITY CERTIFICATION

The need of stable, homogeneous and distinct describers which can be used in
certification and identification of bean cultivars, increased significantly with cultivar protection
and register law. With the aim of studying markers for genetic purity certification of carioca bean
seeds the characterization of six cultivars of bean were realized, Carioca, Carioca MG, Aporé,
Pérola, IAPAR 57 and IAPAR 81 by genetic similarity study. The project was conducted at
experimental area and seed analysis laboratory at the Federal University of Lavras. Electrophoretic
patterns of storage proteins obtained by SDS-PAGE showed similarities between Aporé and Pérola
cultivars. With the data obtained by isoelectric focusing was estimated the genetic similarity by
Jaccard coeficient and the cultivars separated in two groups: I) Carioca, Carioca MG and IAPAR
57; II) Aporé, Pérola and IAPAR 81. The enzymatic systems peroxidase, gluthamate oxilacethate
transaminase, diaphorase, sterase and alchool desidrogenase were studied by Native-PAGE,
estimated the genetic similarity by Jaccard coeficient and the cultivars separeted in four groups: I)
Carioca and Carioca MG; II) Pérola; III) IAPAR 81; IV) Aporé and IAPAR 57. Sterase and
diaphorase discriminated Aporé and Carioca cultivars respectively. Alchool desidrogenase
discriminated the cultivars Carioca and Carioca MG, Aporé and IAPAR 57, Pérola and IAPAR 81.
