PREPARATION AND COLORATION OF Albizzia hasslerii (Chodat) Burr. SEEDS FOR THE TETRAZOLIUM TEST EVALUATION

Procedures to tetrazolium test standartization were conducted to evaluate Albizia
hasslerii
(Chodat) Burr. seed quality. Seeds of that especies were submitted to the following
preparations: control; seeds without seed coat extraction soaked in water for 24 hours; water
soaked for 24 hours followed by seed coat removal; water soaked for 24 hours followed by seed
coat removal and longitudinal cut through embryo axis; and manual scarification with sandpaper
followed by water imbibition for 24 hours. All the pre-conditioning were exposed for three and
five hours, in solution of 2,3,5 triphenyltetrazolium-chloride with concentration of 0.1 and 0.2%.
The treatments with water soaked for 24 hours, followed by seed coat extraction and manual
scarification with sandpaper, followed by water imbibition showed uniform coloration and with
adequate intensity. The treatments with water soaked for 24 hours, followed by seed coat extraction
in solution with concentration of 0.1% for five hours of coloration presented adequate for tetrazolium
test.
